About My Project

Secondary structure, and consequently the folding state of a protein, plays a major role in its function as it is the key element in the prediction of tertiary structure and also useful in assessing protein aggregation and stability. The use and combination of several spectroscopic techniques will serve for the purpose of estimating the protein motif of various biopharmaceutical proteins.

Highly sensitive optical measurements, such as CD (circular dichroism) (Fig. 1), IR and Raman spectroscopies are quite rapid and require very little material, thus contributing to the concept of Quality by Design. In addition to this, high-resolution 1H-NMR (Fig. 2) spectroscopy can provide us with a rough idea about the secondary structure and the stability of a protein, while 2D and nD NMR provides more detailed information about the structure and dynamics of proteins at the atomic level. The combination of NMR with the previously mentioned low-resolution methods will help to bridge the gap between amino acid sequence and function by providing clues to the structure a protein may adopt.
